What is Tirzepatide?

Tirzepatide is a novel multifunctional dual incretin mimetic designed based on the native GIP peptide sequence with amino acid modifications and a C20 diacid moiety attached via a hydrophilic linker. In the SURPASS and SURMOUNT clinical trials, Tirzepatide demonstrated unprecedented reductions in glycated hemoglobin (HbA1c) and visceral adipose mass.

Biological Mechanism

Acts simultaneously on both GIP and GLP-1 receptors. The GIP receptor activity promotes insulin secretion and enhances lipid buffering capacity in subcutaneous adipose tissue while dampening nausea, while the GLP-1 component suppresses appetite and slows gastric transit.

Tirzepatide Once Weekly for the Treatment of Obesity (SURMOUNT-1)

Multinational phase 3 trial establishing substantial and sustained body-weight reductions of up to 20.9% with dual GIP/GLP-1 receptor co-agonist tirzepatide.

Primary Result: Up to -20.9% mean weight loss at 72 weeks at highest dose

Tirzepatide versus Semaglutide Once Weekly in Patients with Type 2 Diabetes (SURPASS-2)

Head-to-head phase 3 trial demonstrating that the dual GIP/GLP-1 receptor co-agonist tirzepatide achieved superior glycemic control and weight reduction compared to semaglutide 1 mg.

Primary Result: -2.37% HbA1c reduction (15 mg tirzepatide) vs. -1.86% (1 mg semaglutide)
