What is Tesamorelin?

Tesamorelin (trade name Egrifta) incorporates the full 44-amino acid sequence of human GHRH stabilized by a hydrophobic trans-hexenoyl fatty acid moiety at the N-terminus. This structural addition imparts superior enzymatic resistance to DPP-4 cleavage compared to native GHRH (1-44). In randomized trials, Tesamorelin demonstrated significant selective reduction in visceral adipose tissue (VAT) and hepatic steatosis.

Biological Mechanism

Binds with high affinity to pituitary GHRH receptors, stimulating pulsatile synthesis and release of endogenous growth hormone and hepatic IGF-1, which selectively promotes lipolysis in visceral fat depots while preserving subcutaneous adipose.
